The Story

50X TAE Buffer (Tris-Acetate-EDTA) is a concentrated stock solution widely used for DNA and RNA electrophoresis. It can be easily diluted to 1X or 0.5X working strength depending on the needs of the experiment.

Tris helps maintain a stable pH during electrophoresis, acetate provides the counter ion for current flow, and EDTA binds divalent cations like Mg²⁺, protecting nucleic acids from nuclease activity.

Formulation (1X working solution): 40 mM Tris, 20 mM Acetic acid, 1 mM EDTA, prepared with ultrapure 18.2 MΩ water.
